Small molecule therapeutics represent the majority of the FDA-approved drugs. Yet, many attractive targets are poorly tractable by small molecules, generating a need for new therapeutic modalities. Due to their biocompatibility profile and structural versatility, peptide-based therapeutics are a possible solution. Additionally, in the past two decades, advances in peptide design, delivery, formulation, and devices have occurred, making therapeutic peptides an attractive modality. However, peptide manufacturing is often limited to solid-phase peptide synthesis (SPPS), liquid phase peptide synthesis (LPPS), and to a lesser extent hybrid SPPS/LPPS, with SPPS emerging as a predominant platform technology for peptide synthesis. SPPS involves the use of excess solvents and reagents which negatively impact the environment, thus highlighting the need for newer technologies to reduce the environmental footprint. Herein, fourteen American Chemical Society Green Chemistry Institute Pharmaceutical Roundtable (ACS GCIPR) member companies with peptide-based therapeutics in their portfolio have…

A visible-light-induced chemodivergent synthesis of tetracyclic quinazolinones and 3-iminoisoindoliones has been developed. This chemodivergent reaction afforded two kinds of different products by substrate control. A detailed investigation of the reaction mechanism revealed that this consecutive photoinduced electron transfer (ConPET) cascade cyclization involved a radical process, and the aryl radical was the crucial intermediate. This method employed 4-DPAIPN as a photocatalyst and i -Pr 2 NEt as a sacrificial electron donor leading to metal-free conditions.

The release of strain energy is a fundamental driving force for organic reactions. However, absolute strain energy alone is an insufficient predictor of reactivity, evidenced by the similar ring strain but disparate reactivity of cyclopropanes and cyclobutanes. In this work, we demonstrate that electronic delocalization is a key factor that operates alongside strain release to boost, or even dominate, reactivity. This delocalization principle extends across a wide range of molecules containing three-membered rings such as epoxides, aziridines, and propellanes and also applies to strain-driven cycloaddition reactions. Our findings lead to a "rule of thumb" for the accurate prediction of activation barriers in such systems, which can be easily applied to reactions involving many of the strained building blocks commonly encountered in organic synthesis, medicinal chemistry, polymer science, and bioconjugation. Given the significance of electronic delocalization in organic chemistry, for example in aromatic π-systems and hyperconjugation, we anticipate that this concept will serve as a…

Organocatalytic desymmetrization reaction is a powerful tool for constructing axial chirality, but the theoretical study on the origin of stereoselectivity still lags behind even now. In this work, the N-heterocyclic carbene (NHC)-catalyzed desymmetrization reaction of biaryl frameworks for the synthesis of axially chiral aldehydes has been selected and theoretically investigated by using density functional theory (DFT). The fundamental pathway involves several steps, i.e., desymmetrization, formation of Breslow oxidation, esterification, and NHC regeneration. The desymmetrization and formation of Breslow processes have been identified as stereoselectivity-determining and rate-determining steps. Further weak interaction analyses proved that the C-H···O hydrogen bond and C-H···π interactions are responsible for the stability of the key stereoselective desymmetrization transition states. This research contributes to understanding the nature of NHC-catalyzed desymmetrization reactions for the synthesis of axially chiral compounds.

Presented herein are novel syntheses of CF 3 -isoquinolinones and imidazole fused CF 3 -isoquinolinones based on the cascade reactions of 2-aryloxazolines with trifluoromethyl imidoyl sulfoxonium ylides. The formation of CF 3 -isoquinolinone involves an intriguing cascade process including oxazolinyl group-assisted aryl alkylation through C(sp 2 )–H bond metalation, carbene formation, migratory insertion, and proto-demetalation followed by intramolecular condensation and water-promoted oxazolinyl ring-scission. With this method, the isoquinolinone scaffold tethered with valuable functional groups was effectively constructed. By taking advantage of the functional groups embedded therein, the products thus obtained could be readily transformed into imidazole-fused CF 3 -isoquinolinones or coupled with some clinical drugs to furnish hybrid compounds with potential applications in drug development. In general, the developed protocols feature expeditious and convenient formation of valuable CF 3 -heterocyclic skeletons, broad substrate scope, and ready scalability. In addition, studies…

In the last several years, atropisomers owing to the rotational restriction around a C-N single bond (C-N axially chiral compounds) have attracted significant attention in the field of synthetic organic chemistry. In particular, the highly enantioselective synthesis of various C-N axially chiral compounds and their application to asymmetric reactions have been reported by many groups. On the other hand, studies on the structural chemistry of C-N axially chiral compounds have attracted scant attention in comparison with synthetic studies. For over 25 years, our group has explored asymmetric synthesis of C-N axially chiral compounds and their synthetic application. In the course of these synthetic studies, we found several notable structural properties in relation to the C-N bond rotation and an association of enantiomers (the relationship between the rotational stability and the structure or electronic effect, the chirality-dependent halogen bond, and the self-disproportionation of enantiomers). Furthermore, on the basis of these structural properties, the development of…

Herein, we demonstrate the direct aziridination of alkenes by combining sulfonyl azides as a triplet nitrene source with a catalytic amount of an organic dye functioning as photosensitizer. We show how the nature of the sulfonyl azide, in combination with the triplet-excited state energy of the photosensitizer, affects the aziridination yield and provide a mechanistic rationale to account for the observed dependence of the reaction yield on the nature of the organic dye and sulfonyl azide reagents. The development of one-pot, atom, and step-economic new methods avoiding metal, harsh reaction conditions, and toxic reagents for the synthesis of medicinally important hybrid molecules bearing more than one bioactive moieties is currently one of the hot topics in organic synthesis. Herein, we report a green and efficient room temperature multicomponent reaction for the synthesis of novel pyrazole-linked thiazoles involving a one-pot C-C, C-N, and C-S bond-forming process from the reaction of aryl glyoxal, aryl thioamide, and pyrazolones in 1,1,1,3,3,3-hexafluoroisopropanol, a hydrogen bond donating reaction medium. A set of diverse hybrid molecules bearing thiazole and pyrazole moieties were prepared in good to excellent yields by using this method. This methodology can also be extended to prepare thiazole-linked barbiturates as well as imidazole-linked pyrazoles. All the products were fully characterized by spectroscopic techniques. The notable features of this protocol are room temperature, metal as well as additive-free reaction conditions, use of recyclable solvent, water as…

The site-selective functionalization of aromatic compounds via C–H activation has emerged as a popular tool in organic synthesis. In this study, we report a regioselective coupling of maleimide to 2-arylbenzo[ d ]thiazoles in the presence of a rhodium(III) catalyst. Depending upon the nature of the substituent ( R 2 -group) present in the maleimide substrate, either mono- or bis-1,4-addition products were observed in this methodology. In the case of R 2 = aryl, cyclohexyl, and tert -butyl, mono coupling was observed, whereas substituents, such as methyl, ethyl, benzyl, and methyl thiophene, provided bis coupling as the major products. Similar selectivity was also observed in the case of 2-arylbenzo[ d ]oxazoles.

A novel iodine-promoted difunctionalization of α-C sites in enaminones was demonstrated as a means of synthesizing a variety of fully substituted thiazoles by constructing C-C(CO), C-S, and C-N bonds. This transformation allows the realization of enaminones as unusual aryl C2 synthons and simultaneously allows the thioylation and dicarbonylation of α-C sites. A preliminary mechanistic study was performed and indicated that the cleavage of C═C bonds in enaminones involves a bicyclization/ring-opening and oxidative coupling sequence.

Herein, employing well-defined redox noninnocent cobalt(II) complexes an efficient sp 3 C–H alkylation of fluorenes using alcohols as alkylating agents to result in alkylated fluorenes is reported. The catalytic protocol was versatile with various fluorenes and benzyl alcohols. It also showed very good functional group tolerance with both alcohols and fluorenes. Moreover, an efficient single-step and simultaneous di C–C as well as both C–C and the C–N alkylation reaction of fluorenes was observed with this catalytic protocol. Such selective single-step dialkylation of fluorenes is indeed beneficial. Several control experiments, deuterium labeling, and 1 H NMR kinetic studies have revealed a ligand radical-based borrowing hydrogen mechanism involving the azo-aromatic complexes of cobalt as catalysts for the alkylation of fluorenes.

A tetraphenyl imidazole-appended benzothiadiazole-based fluorogenic probe (4,7-bis(4-(1,4,5-triphenyl-1 H -imidazol-2-yl)phenyl)benzo[ c ][1,2,5]thiadiazole ( BIPT )) has been successfully synthesized and characterized by NMR and high-resolution mass spectrometry (HRMS) spectral analyses. A low limit of detection (LOD) can be achieved to detect picric acid (PA; 7.89 nM). When benzothiadiazole acceptors are incorporated in the D–A–D probe, it can produce a large Stokes shift (206 nm) as a result. Fascinatingly, the fluorescence signals of BIPT were ratiometrically induced by the interaction with PA and exhibited an apparent emission shift from pink to green. The detection process of BIPT is triggered by an intermolecular charge transfer process, as the charge transfer takes place from the electron-rich imidazole to the electron-deficient PA. Moreover, fluorescence detection of PA has been employed in paper strips. Advantageously, sensor BIPT can potentially be applied to contact mode and real-time detection of PA in environmental water samples. Additionally, the BIPT sensor has been…

Carbon monoxide (CO) is notorious for its toxic effects but is also recognized as a gasotransmitter with considerable therapeutic potential. Due to the inherent challenges in its delivery, the utilization of organic CO photoreleasing molecules (photoCORMs) represents an interesting alternative to CO administration characterized by high spatial and temporal precision of release. This paper focused on the design, synthesis, and photophysical and photochemical studies of 20 3-hydroxyflavone (flavonol) and 3-hydroxyflavothione derivatives as photoCORMs. Newly synthesized compounds bearing various electron-donating and electron-withdrawing groups show bathochromically shifted absorption maxima and considerably enhanced CO release yields compared to the parent unsubstituted flavonol, exceeding 0.8 equiv of released CO in derivatives exhibiting excited states with a charge-transfer character. Until now, such outcomes have been limited to flavonol derivatives possessing a π-extended aromatic system. In addition, thione analogs of flavonols, 3-hydroxyflavothiones, show substantial…

The reaction afforded a series of tri/difluoromethylated pyrrolo[1,2- a ]indoles in moderate to good yields. The DFT study suggests that the reaction is ascribed to a rhodamine 6G-induced cyclization cascade involving vinyl addition-radical relay and hydrogen-atom-abstraction (HAA) processes, and interestingly, pyrrolo[1,2- a ]indoles are applied as fluorescent dyes into the fluorescence spectrum and live-cell imaging. This paper represents an initial example on photocatalytic cyclization cascades by radical relay and the HAA process.
